    The 'Goldilocks Zone': getting the measure of manual asymmetries

    Some studies have shown that manual asymmetries decrease in older age. These results have often been explained with reference to models of reduced hemispheric specialisation. An alternative explanation, however, is that hand differences are subtle, and capturing them requires tasks that yield optimal performance with both hands. Whereas the hemispheric specialisation account implies that reduced manual asymmetries should be reliably observed in older adults, the ‘measurement difficulty’ account suggests that manual asymmetries will be hard to detect unless a task has just the right level of difficulty – i.e. within the ‘Goldilocks Zone’, where it is not too easy or too hard, but just right. Experiment One tested this hypothesis and found that manual asymmetries were only detected when participants performed in this zone; specifically, performance on a tracing task was only superior in the preferred hand when task constraints were high (i.e. fast speed tracing). Experiment Two used three different tasks to examine age differences in manual asymmetries; one task produced no asymmetries, whilst two tasks revealed asymmetries in both younger and older groups (with poorer overall performance in the old group across all tasks). Experiment Three revealed task-dependent asymmetries in both age groups, but highlighted further detection difficulties linked with the metric of performance and compensatory strategies used by participants. Results are discussed with reference to structural learning theory, whereby we suggest that the processes of inter-manual transfer lead to relatively small performance differences between the hands (despite a strong phenomenological sense of performance disparities)

    Economic evidence for the prevention and treatment of atopic eczema: a protocol for a systematic review

    Background: Eczema, synonymous with atopic eczema or atopic dermatitis, is a chronic skin disease that has a similar impact on health-related quality of life as other chronic diseases. The proposed research aims to provide a comprehensive systematic assessment of the economic evidence base available to inform economic modelling and decision making on interventions to prevent and treat eczema at any stage of the life course. Whilst the Global Resource of Eczema Trials (GREAT) database collects together the effectiveness evidence for eczema there is currently no such systematic resource on the economics of eczema. It is important to gain an overview of the current state of the art of economic methods in the field of eczema in order to strengthen the economic evidence base further. Methods/design: The proposed study is a systematic review of the economic evidence surrounding interventions for the prevention and treatment of eczema. Relevant search terms will be used to search MEDLINE, EMBASE, Database of Abstracts of Reviews of Effects, Cochrane Database of Systematic Reviews, Cochrane Central Register of Controlled Trials, NHS Economic Evaluation Database, Health Technology Assessment, Cumulative Index to Nursing and Allied Health Literature, Econ Lit, Scopus, Cost-Effectiveness Analysis Registry and Web of Science in order to identify relevant evidence. To be eligible for inclusion studies will be primary empirical studies evaluating the cost, utility or full economic evaluation of interventions for preventing or treating eczema. Two reviewers will independently assess studies for eligibility and perform data abstraction. Evidence tables will be produced presenting details of study characteristics, costing methods, outcome methods and quality assessment. The methodological quality of studies will be assessed using accepted checklists. Discussion: The systematic review is being undertaken to identify the type of economic evidence available, summarise the results of the available economic evidence and critically appraise the quality of economic evidence currently available to inform future economic modelling and resource allocation decisions about interventions to prevent or treat eczema. We aim to use the review to offer guidance about how to gather economic evidence in studies of eczema and/or what further research is necessary in order to inform this

    Valproic acid protects against haemorrhagic shock-induced signalling changes via PPARγ activation in an in vitro model.

    BACKGROUND AND PURPOSE: Valproic acid (VPA), a widely used epilepsy and bipolar disorder treatment, provides acute protection against haemorrhagic shock-induced mortality in a range of in vivo models through an unknown mechanism. In the liver, this effect occurs with a concomitant protection against a decrease in GSK3β-Ser(9) phosphorylation. Here, we developed an in vitro model to investigate this protective effect of VPA and define a molecular mechanism. EXPERIMENTAL APPROACH: The human hepatocarcinoma cell line (Huh7) was exposed to conditions occurring during haemorrhagic shock (hypoxia, hypercapnia and hypothermia) to investigate the changes in GSK3β-Ser(9) phosphorylation for a 4 h period following treatment with VPA, related congeners, PPAR agonists, antagonists and siRNA. KEY RESULTS: Huh7 cells undergoing combined hypoxia, hypercapnia, and hypothermia reproduced the reduced GSK3β-Ser(9) phosphorylation shown in vivo during haemorrhagic shock, and this change was blocked by VPA. The protective effect occurred through upstream PTEN and Akt signalling, and prevented downstream β-catenin degradation while increasing histone 2/3 acetylation. This effect was reproduced by several VPA-related compounds with known PPARγ agonist activity, independent of histone deacetylase (HDAC) inhibitory activity. Specific pharmacological inhibition (by T0070907) or knockdown of PPARγ blocked the protective effect of VPA against these signalling changes and apoptosis. In addition, specific activation of PPARγ using ciglitazone reproduced the changes induced by VPA in haemorrhagic shock-like conditions. CONCLUSION AND IMPLICATIONS: Changes in GSK3β-Ser(9) phosphorylation in in vivo haemorrhagic shock models can be modelled in vitro, and this has identified a role for PPARγ activation in the protective role of VPA

    Do PTK2 gene polymorphisms contribute to the interindividual variability in muscle strength and the response to resistance training? A preliminary report.

    The protein tyrosine kinase-2 (PTK2) gene encodes focal adhesion kinase, a structural protein involved in lateral transmission of muscle fiber force. We investigated whether single-nucleotide polymorphisms (SNPs) of the PTK2 gene were associated with various indexes of human skeletal muscle strength and the interindividual variability in the strength responses to resistance training. We determined unilateral knee extension single repetition maximum (1-RM), maximum isometric voluntary contraction (MVC) knee joint torque, and quadriceps femoris muscle specific force (maximum force per unit physiological cross-sectional area) before and after 9 wk of knee extension resistance training in 51 untrained young men. All participants were genotyped for the PTK2 intronic rs7843014 A/C and 3'-untranslated region (UTR) rs7460 A/T SNPs. There were no genotype associations with baseline measures or posttraining changes in 1-RM or MVC. Although the training-induced increase in specific force was similar for all PTK2 genotypes, baseline specific force was higher in PTK2 rs7843014 AA and rs7460 TT homozygotes than in the respective rs7843014 C- (P = 0.016) and rs7460 A-allele (P = 0.009) carriers. These associations between muscle specific force and PTK2 SNPs suggest that interindividual differences exist in the way force is transmitted from the muscle fibers to the tendon. Therefore, our results demonstrate for the first time the impact of genetic variation on the intrinsic strength of human skeletal muscle

    Before the whistle blows: developing new paradigms in tuberculosis screening to maximise benefit and minimise harm [version 1; peer review: awaiting peer review]

    We summarise recent emerging evidence around tuberculosis (TB) transmission and its role in tuberculosis epidemiology, and in novel TB screening and diagnostic tests that will likely become available in low-resource settings in the near future. Little consideration has been paid to how these novel new tests will be implemented, nor what the consequences for individuals, communities and health systems will be. In particular, because of low specificity and consequent false-positive diagnoses, and the low percentage of people who “screen positive” that will go onto develop active pulmonary disease, there is significant potential for inappropriate initiation of TB treatment, as well as stigmatisation, loss of livelihoods and in some setting institutionalisation, with uncertain benefit for individual health or community transmission. We use analogy to prompt consideration of how and where new TB screening tests could be implemented in TB screening programmes in low-resource settings. Acceptance and confidence in TB screening programmes depends on well-functioning public health programmes that use screening algorithms that minimise harms and balance population benefits with autonomy and respect for individuals. Before new TB screening tests and algorithms are introduced, more evidence for their effectiveness, costs, benefits and harms under real-world conditions are required

    In the beginning: Role of autonomy support on the motivation, mental health and intentions of participants entering an exercise referral scheme

    Self-determination theory (Deci & Ryan, 2000, Intrinsic motivation and self-determination in human behaviour. New York: Plenum Publishing) highlights the impact autonomy supportive environments can have on exercise motivation and positive health outcomes. Yet little is known about whether differential effects occur as a function of which significant other is providing this support. Further, no research has examined the relationship between motivation and the social environment with participants’ mental health and intentions to be physically active before entering an exercise intervention. Study participants were 347 British adults who were about to start an exercise referral scheme. Regression analyses revealed that the effects of autonomy support on mental health and physical activity intentions differed as a function of who provided the support (offspring, partner or physician), with the offspring having the weakest effects. A structural model was supported, indicating that autonomy support and more autonomous regulations led to more positive mental health outcomes and stronger intentions to be physically active. Knowledge of the social environmental and personal motivation of those about to commence an exercise programme can provide important insights for professionals supporting such efforts

    Net Zero requires ambitious greenhouse gas emission reductions on beef and sheep farms coordinated with afforestation and other land use change measures

    CONTEXT: The UK Climate Change Committee has recommended a 64% reduction in greenhouse gas emissionsfrom the agriculture and land-use sector to meet the 2050 Net Zero target in the UK. However, it is unclear howthis reduction can be achieved at a farm level.OBJECTIVE: Using detailed real farm data and novel modelling approaches, we investigated the managementinterventions and afforestation that would be required to deliver Net Zero within the farm boundary.METHODS: Baseline carbon footprints were calculated for twenty Welsh beef and sheep farms using the Agrecalccarbon calculator, whilst carbon sequestration was estimated using Bangor University’s Carbon FootprintingTool. Scenarios were created to determine the emissions reductions achievable on each farm through implementationof cost-effective mitigation measures. Mitigation measures and their abatement potentials weresourced from the most recent UK Marginal Abatement Cost Curve, which allow emissions to be reduced mostlythrough improvements in efficiency thus maintaining the production of the system. Area footprints werecalculated for production, with and without offset (afforested) areas needed to achieve Net Zero

    Multi-robot Collaborative Visual Navigation with Micro Aerial Vehicles

    Micro Aerial Vehicles (MAVs), particularly multi-rotor MAVs have gained significant popularity in the autonomous robotics research field. The small size and agility of these aircraft makes them safe to use in contained environments. As such MAVs have numerous applications with respect to both the commercial and research fields, such as Search and Rescue (SaR), surveillance, inspection and aerial mapping. In order for an autonomous MAV to safely and reliably navigate within a given environment the control system must be able to determine the state of the aircraft at any given moment. The state consists of a number of extrinsic variables such as the position, velocity and attitude of the MAV. The most common approach for outdoor operations is the Global Positioning System (GPS). While GPS has been widely used for long range navigation in open environments, its performance degrades significantly in constrained environments and is unusable indoors. As a result state estimation for MAVs in such constrained environments is a popular and exciting research area. Many successful solutions have been developed using laser-range finder sensors. These sensors provide very accurate measurements at the cost of increased power and weight requirements. Cameras offer an attractive alternative state estimation sensor; they offer high information content per image coupled with light weight and low power consumption. As a result much recent work has focused on state estimation on MAVs where a camera is the only exteroceptive sensor. Much of this recent work focuses on single MAVs, however it is the author's belief that the full potential and benefits of the MAV platform can only be realised when teams of MAVs are able to cooperatively perform tasks such as SaR or mapping. Therefore the work presented in this thesis focuses on the problem of vision-based navigation for MAVs from a multi-robot perspective. Multi-robot visual navigation presents a number of challenges, as not only must the MAVs be able to estimate their state from visual observations of the environment but they must also be able to share the information they gain about their environment with other members of the team in a meaningful fashion. The meaningful sharing of observations is achieved when the MAVs have a common frame of reference for both positioning and observations. Such meaningful information sharing is key to achieving cooperative multi-robot navigation. In this thesis two main ideas are explored to address these issues. Firstly the idea of appearance based (re)-localisation is explored as a means of establishing a common reference frame for multiple MAVs. This approach allows a team of MAVs to very easily establish a common frame of reference prior to starting their mission. The common reference frame allows all subsequent operations, such as surveillance or mapping, to proceed with direct cooperative between all MAVs. The second idea focuses on the structure and nature of the inter-robot communication with respect to visual navigation; the thesis explores how a partially distributed architecture can be used to vastly improve the scalability and robustness of a multi-MAV visual navigation framework. A navigation framework would not be complete without a means of control. In the multi-robot setting the control problem is complicated by the need for inter-robot collision avoidance. This thesis presents a MAV trajectory controller based on a combination of classical control theory and distributed Velocity Obstacle (VO) based collision avoidance. Once a means of control is established an autonomous multi-MAV team requires a mission. One such mission is the task of exploration; that is exploration of a previously unknown environment in order to produce a map and/or search for objects of interest. This thesis also addressed the problem of multi-robot exploration using only the sparse interest-point data collected from the visual navigation system. In a multi-MAV exploration scenario the problem of task allocation, assigning areas to each MAV to explore, can be a challenging one. An auction-based protocol is considered to address the task allocation problem. The two applications discussed, VO-based trajectory control and auction-based environment exploration, form two case studies which serve as the partial basis of the evaluation of the navigation solutions presented in this thesis. In summary the visual navigation systems presented in this thesis allow MAVs to cooperatively perform task such as collision avoidance and environment exploration in a robust and efficient manner, with large teams of MAVs. The work presented is a step in the direction of fully autonomous teams of MAVs performing complex, dangerous and useful tasks in the real world
